Functional expression and purification of tailor-made chimeric endolysin with the broad antibacterial spectrum

摘要: Developing chimeric lysins with a wide lytic spectrum is important in fighting against Gram-negative pathogenic bacteria. In the present work, novel chimerical lysin, LytAmfi, was constructed by fusing bacteriophage endolysin Lyt μ1/6 an amphipathic cationic peptide derived from T4 lysozyme. The result showed that LytAmfi had not only activity similar to parental Streptomyces aureofaciens but also broadened strains. Our work demonstrated generating lysin extended workable active modular could penetrate outer membrane of bacteria including Escherichia coli, Pseudomonas agglomerans, Acinetobacter lwoffii, Hafnia alvei, Citrobacter freundii and enable access lyse ‘from within’ host cell without pre-treatment its membrane. Therefore, would be appreciable therapeutic value.
