摘要: Abstract This study confirms that normal human articular chondrocytes express neutrophil collagenase or matrix metalloproteinase-8 (MMP-8), a gene product previously thought to be expressed exclusively by leukocytes. Both MMP-8 protein and mRNA were present in cartilages collected from donors. Cartilage extracts assayed immunoblotting analysis of enzymatic activity on gelatin-substrate gels. Latent extracted cartilage has molecular mass 55 kDa; active was identified at 46 42 kDa. In the absence reducing agent, migrated high complex above 200 Northern blotting results demonstrated expression chondrocytes, which could up-regulated stimulation with interleukin-1β. addition, reverse transcription-polymerase chain reaction using nested primers situ hybridization revealed presence chondrocytes. The both message supports concept enzyme described as “aggrecanase.”
