The antibacterial and antifungal properties of trappin-2 (pre-elafin) do not depend on its protease inhibitory function.

摘要: Trappin-2 (also known as pre-elafin) is an endogenous inhibitor of neutrophil serine proteases and involved in the control excess proteolysis, especially inflammatory events, along with structurally related secretory leucocyte proteinase inhibitor. Secretory has been shown to have antibacterial antifungal properties, whereas recent data indicate that trappin-2 antimicrobial activity against Pseudomonas aeruginosa Staphylococcus aureus. In present study, we tested properties towards other respiratory pathogens. We found trappin-2, at concentrations 5-20 microm, significant Klebsiella pneumoniae, Haemophilus influenzae, Streptococcus Branhamella catarrhalis pathogenic fungi Aspergillus fumigatus Candida albicans, addition P. S. A similar was observed A62D/M63L, a variant lost its antiprotease indicating exerts effects through mechanisms independent from intrinsic capacity. Furthermore, activities were sensitive NaCl heparin, demonstrating mechanism action most probably dependent on cationic nature. This enables interact membranes target organisms disrupt them, by our scanning electron microscopy analyses. Thus, not only provides shield, but also may play important role innate defense human lungs mucosae microorganisms.
