Loss of the major Type I arginine methyltransferase PRMT1 causes substrate scavenging by other PRMTs

摘要: Arginine methylation is a common posttranslational modification that found on both histone and non-histone proteins. Three types of arginine exist in mammalian cells: monomethylarginine (MMA), asymmetric dimethylarginine (ADMA) symmetric (SDMA). PRMT1 the primary methyltransferase deposits ADMA mark, it accounts for over 90% this type methylation. Here, we show with loss activity, there are major increases global MMA SDMA levels, as detected by type-specific antibodies. Amino acid analysis confirms levels accumulate when reduced. These findings reveal dynamic interplay between different cells, pre-existence dominant mark can block occurrence marks same substrate. This study provides clear evidence competition substrates.
