摘要: In cells of Old World and some New monkeys, dominant factors restrict human immunodeficiency virus type 1 (HIV-1) infections after entry. The simian virus SIV mac is less susceptible to these restrictions, a property that determined largely by the viral capsid protein. For this study, we altered exposed amino acid residues on surface HIV-1 capsid, changing them corresponding found the SIV capsid. We identified two distinct pathways escape from early, postentry restriction in monkey cells. One set mutants that were near base cyclophilin A-binding loop of the N-terminal domain or interdomain linker exhibited a decreased ability bind restricting factor(s). Consistent with the location putative factor-binding site, A and the factor(s) cooperated achieve block. A second mutants that ridge formed helices 3 6 efficiently bound the restricting but resistant consequences factor binding. These results imply binding restricting factor(s) not sufficient mediate block HIV-1 and that SIV capsids escape decreases both factor susceptibility effects the factor(s).
