Kinetics and metabolism of carbamazepine during combined antiepileptic drug therapy

摘要: The kinetics of carbamazepine using 15N-carbamazepine were investigated in epileptic patients during combined anticonvulsant therapy. plasma half-lives ranged from 5.0 to 13.6 hr with a mean 8.2 hr. These are appreciably shorter than reported chronic monotherapy. Predicted steady-state levels and observed excellent agreement. Between 32% 61% the dose administered is excreted urine as carbamazepine-trans-diol, 5.2% 8.8% 9-hydroxymethyl-10-carbamoyl acridane, 1% 1.4% 10,-11-carbamazepine epoxide, 0.5% carbamazepine. data indicate that it epoxide-diol pathway which induced long-term treatment. Concomitant therapy primidone, phenytoin, phenobarbital, ethosuximide, or methsuximide further induces metabolism.
