摘要: The initiation of DNA replication is a key event in the cell cycle all organisms. In bacteria, occurs at specific origin sequences that are recognized and processed by an oligomeric complex initiator protein DnaA. We have determined structure conserved core Aquifex aeolicus DnaA to 2.7 A resolution. comprises AAA+ nucleotide-binding fold linked through long, helical connector all-helical DNA-binding domain. serves as template for understanding physical consequences variety mutations, motifs suggest how two critical aspects processing, binding homo-oligomerization, mediated. spatial arrangement these similar eukaryotic-like archaeal factor Cdc6/Orc1, demonstrating mechanistic elements processing may be across bacterial, eukaryotic domains life.
