Basophils (Bsp-1+) derive from the leukemic clone in human myeloid leukemias involving the chromosome breakpoint 9q34.

摘要: The monoclonal antibody (MoAb) Bsp-1 was used to purify basophilic cells from leukemic blood of five patients with Philadelphia chromosome (Ph′) positive chronic myeloid leukemia (CML) and two acute (AML) characterized by the chromosomal translocation t(6;9)(p23;q34). When cultured, all CML AML showed same clonal karyotype changes observed in diagnostic buffy coat preparations, indicating that were origin. In contrast, T lymphocytes four cultured presence interleukin- 2 (IL-2) a normal therefore not derived clone. staining correlated toluidine blue- basophils phase negative blast expressing an immature phenotype crisis AML. Chromosome situ hybridization ABL oncogene translocated 9 22 but remained on patients. These results indicate breakpoint at 9q34 is 5′ ABL, whereas 3′ ABL. Field inversion gel electrophoresis within 200 kb one patients, nor there any rearrangement PIM locus 6p21.
