摘要: A number of studies have clearly demonstrated that flagellin is a potent adjuvant promotes robust immune responses when it given with protein antigen. In view the potential biological and practical benefits recombinant vaccine composed single fusion containing antigen, we evaluated efficacy two protective antigens Yersinia pestis (F1 V) in eliciting protection against respiratory challenge Y. pestis. Flagellin-F1-V was produced purified high yield under good manufacturing practices conditions. The retains full Toll-like receptor 5-stimulating activity vitro. Using prime-boost immunization protocol, found flagellin-F1-V elicits antigen-specific humoral immunity mice species nonhuman primates. Immune were fully protected intranasal 150 mean tolerated doses CO92. mice, bacteria completely cleared within 3 days after challenge. exhibited stability for at least 297 4°C 168 25°C. At between 29 84 37°C, loss appeared to be associated substantial change diameter, possibly due oligomerization. On basis our results, believe an outstanding candidate evaluation humans.
