摘要: Beta-Galactosidase (EC 3.2.1.23) has been purified from the livers of C57BL/6J mice. The enzyme migrated as a single band protein on polyacrylamide gel electrophoresis in presence sodium dodecyl sulfate. molecular weight denatured and reduced was 63,000. native form beta-galactosidase appeared to be tetramer 240,000 at pH 5.0, which reversibly dissociated alkaline dimer with apparent 113,000. Multiple charge isomers were resolved by ion exchange chromatography. Treatment neuraminidase markedly its electrophoretic mobility. Purified well crude liver extract hydrolyzed p-nitrophenyl-beta-D-fucoside one-tenth rate hydrolysis beta-galactoside. Antiserum precipitated major portion activity mouse liver, brain, kidney. This antiserum cross-reacts beta-galactosidases rat Chinese hamster, but not human, porcine, or bovine beta-galactosidase.
