Protective effects of Withania somnifera extract in SOD1G93A mouse model of amyotrophic lateral sclerosis.

摘要: Withania somnifera (WS; commonly known as Ashwagandha or Indian ginseng) is a medicinal plant whose extracts have been in use for centuries various regions of the world rejuvenator. There now growing body evidence documenting neuroprotective functions its purified compounds several models neurodegenerative diseases including amyotrophic lateral sclerosis (ALS). Based on extract's beneficial effect mouse model ALS with TDP-43 proteinopathy, current study was designed to test efficacy another familial ALS. Our results show that administration WS by gavage mice expressing G93A mutant form superoxide dismutase (SOD1) resulted increased longevity, improved motor performance and number neurons lumbar spinal cord. The treatment caused substantial reduction levels misfolded SOD1whereas it enhanced expression cellular chaperons cord SOD1G93A mice. markedly reduced glial activation prevented phosphorylation nuclear factor kappaB (NF-κB). overall immunomodulatory further evidenced changes multiple cytokines/chemokines. also served an autophagy inducer which may be at early stages disease. These suggest might constitute promising therapeutics involvement SOD1.
