摘要: Abstract An immunochemical method has been used to detect quantitatively DNA damage caused by ionizing radiation in germ cells. With this method, strand breaks as well lesions converted into alkaline medium are measured a function of controlled partial unwinding the DNA, time-dependent process starting at each breakage site, followed determination relative amount single-stranded regions use single-strand specific monoclonal antibody. induction and repair different cellular stages spermatogenesis (spermatocytes, round elongated spermatids) hamster were investigated. Germ cells irradiated vitro with 60 Co-γ-rays, doses between 0 5 Gy. A linear dose-response relationship was observed. Spermatocytes spermatids had normal, fast when compared these sites cultured V79 or CHO human lymphocytes. The spermatids, however, showed hardly any repair. Similar results obtained after vivo γ-irradiation hamsters 0, 4, 8 Gy subsequent isolation still detectable 24 h exposure. experiments show substantial differences capacity cell development.
