Regulation of the mitochondrial ATP synthase in intact rat cardiomyocytes.

摘要: The ATP synthase capacity of rat heart myocytes can be measured in sonicated cell suspensions and preparations cultured cardiomyocytes. This procedure allows the rapid measurement mitochondrial function response to changes metabolic status cell. In myocytes, transitions (with no detectable change cellular concentration) accompany a anoxia or electrically stimulated contraction (rise 70%). These are reversed on returning original conditions. Exposure low pH has little effect basal (down values less than 6), but markedly affects levels cells reoxygenation, possibly mimicking seen ischaemic heart. Similar effects freshly prepared these stable more pH-sensitive culture. It is proposed that mitochondria vivo directly regulated at level synthase, regulator protein, naturally occurring inhibitor protein from mitochondria, may responsible for this regulation.
