摘要: In eukaryotes, mitochondrion is an essential organelle which surrounded by a double membrane system, including the outer membrane, intermembrane space and inner membrane. The translocase of mitochondrial (TOM) complex has attracted enormous interest for its role in importing preprotein from cytoplasm into mitochondrion. However, little understood about potential biological function TOM Arabidopsis. aim present study was to investigate how AtTOM40, gene encoding core subunit complex, works As result, we found that lack AtTOM40 disturbed embryo development pattern formation after globular stage, finally caused albino ovules seed abortion at ratio quarter homozygous tom40 plants. Further investigation demonstrated wildly expressed different tissues, especially cotyledons primordium during Arabidopsis embryogenesis. Moreover, confirmed encoded protein localized mitochondrion, observation ultrastructure revealed biogenesis impaired tom40-1 cells. Quantitative real-time PCR utilized determine expression genes proteins mutant embryos, known be involved import, assembly transport proteins, results most expressions were abnormal. Similarly, relevant formation, such as SAM (shoot apical meristem), cotyledon, vascular hypophysis, also affected embryos. Taken together, draw conclusion normal structure participates early through maintaining mitochondria. findings this may provide new insight TOM40 higher
