Enterococcus hirae WEHI01 isolated from a healthy Chinese infant ameliorates the symptoms of type 2 diabetes by elevating the abundance of Lactobacillales in rats

摘要: Enterococcus hirae WEHI01 is a potential probiotic strain isolated from healthy Chinese infant. This has previously been characterized as having cholesterol-lowering and good dairy fermentation performance. In this study, we used rat models with obesity type 2 diabetes mellitus (T2DM) induced by high fat sucrose diet low-dose streptozotocin, respectively, evaluated the effect of E. on glycolipid metabolism, glycolipid-related gene expression, organ histopathology, intestinal flora changes in models. Our results showed that administration 5.0 × 109 cfu for 4 wk decreased serum lipid levels regulated metabolism liver obese rats. Following continuous same concentration to T2DM model another 5 wk, improved glucose tolerance, recovered body weight loss, led significant decreases tumor necrosis factor-α, IL-6, IL-10, total bile acid serum. We also found restored morphology pancreas, kidney, liver, changed composition gut microbiota (i.e., Shannon index, increased Simpson substantially abundance Lactobacillales). Combining model, speculated beneficial effects could be due (1) increase PPARA expression tendency CYP7A1 rats, promoting conversion cholesterol into reducing rats; or (2) change microbial diversity, especially elevated Lactobacillales abundance, which reduced These demonstrated ameliorate rats provide promising rationale further research prevention treatment T2DM.
