摘要: We describe the use of small interfering RNAs (siRNAs) to down-regulate H- and L-ferritin levels in HeLa cells. siRNAs repressed expression about 20% 25% background level both stable transient transfections. cells transfected with cDNAs were analyzed parallel compare effects ferritin up- down-regulation. found that large modifications did not affect iron availability but positively affected cell proliferation rate an iron-independent manner. The down-regulation H-ferritin modified cellular resistance oxidative damage, as expected. In contrast, suppression clones increase made less resistant supplementation chelation. results indicate has no direct on homeostasis cells, while it new, iron-unrelated functions. addition, they suggest function is act buffer.
