摘要: Filaggrin is an important keratin intermediate filament-associated protein of terminally differentiated mammalian epidermis. Its aberrant expression has been implicated in a number keratinizing disorders. We have isolated and sequenced cDNA clone to mouse filaggrin, 1.479 kilobase pairs, which represents less than 10% the full-length mRNA estimated by Northern blot analysis be 17 kilobases long. The delineates 744-base pair repeat. This encodes 248 amino acids or 26,330 Da, almost identical known properties filaggrin size, acid composition, charge. Total genomic DNA gene from cosmid library were found contain super-stoichiometric repeat same size. These data support hypothesis (Haydock, P.V., Dale, B.A. (1986) J. Biol. Chem. 261, 12520-12525) that initially synthesized as polyprotein precursor containing many tandem copies. However, our suggest repeating units are not separated "large linker" peptides suggested these authors. In situ hybridization was used show located precisely over granular layer epidermis, indicating this regulated at transcriptional level for differentiation-specific 1 protein. probes will now permit detailed studies on regulation gene.
