CircLRP6 Regulation of ZEB1 via miR-455 Is Involved in the Epithelial-Mesenchymal Transition During Arsenite-Induced Malignant Transformation of Human Keratinocytes.

摘要: Circular RNAs (circRNAs), a class of noncoding generated from pre-mRNAs, participate in the regulation tumorigenesis. The mechanism for regulation, however, is unclear. Here, to determine whether circRNAs are involved arsenite-induced epithelial-mesenchymal transition (EMT) and malignant transformation human keratinocyte (HaCaT) cells, up-regulation circLRP6 was confirmed arsenite-transformed HaCaT (T-HaCaT) cells. In acted as an microRNA (miR)-455 sponge. For these chronic exposure arsenite caused increase transcription factor ZEB1, which induced EMT. miR-455 suppressed expression ZEB1. Further, T-HaCaT knockdown with siRNA inhibited ZEB1 expression, but cotransfection inhibitor reversed this inhibition. These results suggest that, increases levels, act sponge up-regulate target, subsequently induces EMT, thus promoting transformation. Thus, cells chronically exposed arsenite, via EMT during establish previously unknown carcinogenesis.
