摘要: Background Although the hygiene hypothesis suggests that microbial infections could subvert asthma and thus a product might serve as therapeutic adjuvant for asthma, relationship between bacterial components is complex. Recently, low levels of flagellin, Toll-like receptor (TLR) 5 ligand, have been reported to promote asthma. Objective We show dose flagellin suppresses effect occurs through generating regulatory dendritic cells (rDCs) T (Treg) cells. Methods Ovalbumin (OVA)–induced wild-type TLR5 knockout asthmatic mice were treated intranasally with mixture OVA 10 μg B (FlaB; Vibrio vulnificus ). OVA/FlaB-treated rDCs adoptively transferred OVA-induced Anti-CD25 mAb was used deplete Treg A mixture house dust mite (HDM) FlaB treat HDM-induced Blood CD14 + monocyte-derived from HDM-sensitive patients incubated autologous CD4 Results An OVA/FlaB ameliorated by inhibiting H 1/T 2/T 17 responses in TLR5-dependent manner The adoptive transfer inhibited whereas depletion CD25 eliminated inhibitory effect. A similar observed In FlaB-treated HDM-stimulated 2 while enhancing an IL-10–dependent manner. Conclusion These findings collectively suggest be tolerogenic allergic
