High frequency of natural autoantibodies in normal newborn mice.

摘要: Spleen cells from 6-day-old nonimmunized BALB/c and BALB.B10 mice were fused with the nonsecreting hybridoma cell line Sp2/0. Three hundred eighty-four immunoglobulin-secreting hybrids screened for antibody activity against mouse actin, tubulin, myosin, TNP, peroxidase, renin, DNA, neurofilaments. At least 24 hybridomas in collection (6.25%) exhibited this panel of antigens. Ten these cloned, propagated, corresponding monoclonal IgM protein was isolated ascitic fluids further characterized. four groups specificities identified: 1) one clone reacting TNP only; 2) both actin tubulin; 3) two clones which bound to actin; 4) a fourth group, comprising six other clones, all widespread reactivity TNP. These results indicate: B directed self antigens are activated internal environment recovered consequently by somatic hybridization; found newborn antibodies very similar those previously characterized human natural Ig; frequency binding cytoskeletal proteins is high (at 6.25%).
