摘要: The susceptibility of hematopoietic progenitor cells to infection by human cytomegalovirus (HCMV) was investigated using several strains HCMV, including the recombinant strain RC256. RC256 is derived from laboratory Towne and contains Escherichia coli LacZ gene coding for beta-galactosidase (beta-gal) regulated an early HCMV promoter. Expression allowed detection in individual cells. Clonogeneic bone marrow (BM) progenitors, CD34+ cells, could be infected with would then form normal colonies. By polymerase chain reaction (PCR) amplification DNA, detected both erythroid myeloid activity observed predominantly myelomonocytic lineage. When HCMV-infected progenitors were cocultivated permissive fibroblasts, infectious virus expressing recovered. Although no characteristic cytopathology BM colonies, high cell ratios resulted a moderate inhibition colony formation. Since can harbor weeks through differentiation steps culture, we postulate that vivo these may serve as reservoir latent contribute dissemination.
