摘要: Mammalian oocytes remain dormant in the diplotene stage of prophase I until resumption meiosis characterized by germinal vesicle breakdown (GVBD) following preovulatory gonadotropin stimulation. Based on genome-wide analysis peri-ovulatory DNA microarray to identify paracrine hormone-receptor pairs, we found increases ovarian transcripts for endothelin-1 and endothelin receptor type A (EDNRA) response luteinizing hormone (LH)/human chorionic (hCG) Immunohistochemical analyses demonstrated localization EDNRA granulosa cumulus cells. In cultured follicles, treatment with promoted oocyte GVBD. The stimulatory effect was blocked cotreatment antagonists A, but not related B, receptor. hCG GVBD partially same antagonist. promotion be mediated MAPK/ERK pathway inhibitory G protein. Studies using cumulus-oocyte complexes denuded that actions are Furthermore, intrabursal administration induced follicles. Our findings demonstrate a role induction provide further understanding molecular mechanisms underlying nuclear maturation LH surge.
