摘要: Abstract Substance P (SP) is an 11-amino acid tachykinin-related peptide that has anorexigenic effects in birds and mammals although the central mechanism not well understood. Hence, objective was to identify appetite-associated hypothalamic mechanisms Japanese quail (Coturnix japonica). Seven days post-hatch, were intracerebroventricularly injected with 0, 0.25, 0.5 or 1.0 nmol of SP monitored for 180 min. On a cumulative basis, received consumed less food 90 min post-injection. non-cumulative intake reduced 0.5 nmol-injected at 30 min Water affected. A comprehensive behavior analysis performed, revealing SP-injected chicks displayed feeding pecks locomotion compared vehicle-injected birds. To molecular mechanisms, hypothalamus isolated 1 h post-injection real-time PCR performed measure mRNA. Agouti-related (AgRP) mRNA chicks. Immunohistochemistry used quantify c-Fos-expressing cells nuclei. There more reactive lateral (LH) paraventricular nucleus (PVN) SP- than The LH PVN collected gene expression analysis. Corticotropin-releasing factor (CRF) urotensin 2 (UTS2) mRNAs greater PVN. In LH, CRF receptor sub-type (CRFR2) kappa opioid quail. Thus, induces potent anorexia coincides increased LH-specific CRFR2 UTS2 Future studies will evaluate whether SP-induced are mediated through receptors.
