摘要: Our aim was to search for proteome changes in peripheral blood mononuclear cells (PBMCs) of MDS patients with refractory cytopenia multilineage dysplasia. PBMCs were isolated from a total 12 samples using Histopaque-1077 solution. The proteins fractioned, separated by 2D SDS-PAGE (pI 4-7), and double-stained. proteomes compared statistically processed Progenesis SameSpots; then identified nano-LC-MS/MS. Protein functional association expression profiles analyzed the EnrichNet application SameSpots hierarchical clustering software, respectively. By comparing cytosolic, membrane, nuclear fractions two groups, 178 significantly (P < 0.05, ANOVA) differing spots found, corresponding 139 unique proteins. Data mining Reactome KEGG databases highlighted possible involvement protein alterations apoptosis, proteasome degradation, heat shock action, signal transduction. Western blot analysis revealed underexpression vinculin advanced fragmentation fermitin-3 patients. To best our knowledge, this is first time that have been Vinculin fermitin-3, involved cell adhesion integrin signaling, shown be dysregulated MDS.
