摘要: Publisher Summary This chapter discusses the assay, purification, and properties of branched-chain amino acid aminotransferase. All purification procedures are generally carried out at 4°. buffers used in supplemented with 2 × 10 -3 M 2-mercaptoethanol, 1 EDTA, -5 pyridoxal phosphate. Addition 2-mercaptoethanol considerably activates enzyme. The purified enzyme gives a single spot on agar or acrylamide-gel electrophoresis. Antiserum against provides precipitin line homologous double diffusion test. On ultracentrifugation, formed symmetrical peak. optimal pH is 8.0. activity brain limited to valine, leucine, isoleucine, α -ketoglutarate. antisera heart enzymes inhibit activities both heterologous enzymes. latter inhibited lesser extent.
