Red Fluorescent Organic Nanoparticle Bioprobes: A Photostable Cytoplasmic Stain for Long Term In Vitro and In Vivo Visualization

摘要: Development of efficient fluorescent organic nanoparticles (FONs) for in vitro and vivo bioimaging is one the emerging areas nanobiotechnology. Here, we report synthesis a novel chromophore, composed cytosine moiety connected to modified 2,1,3-benzothiadiazole tag by an aliphatic chain. Self-assembly molecules DMSO/H2O solution led formation solution-stable, red emissive, biocompatible, nontoxic (nano-Cy-Red). These were biocompatible safe all tested biological organisms; staining protocol was also simple rapid. Nano-Cy-Red exhibited ability penetrate into mouse macrophages, cancer cells, yeast retained living cells several generations serving as cytoplasmic stain. Taking advantage this property, exploit nano-Cy-Red dye direct visualization cell migration up 72 h post transplantation zebrafish embryos.
