MNC-RED A Chemically-Defined Method to Produce Enucleated Red Blood Cells from Adult Peripheral Blood Mononuclear Cells

摘要: Abstract Many methods have been developed to produce red blood cells in vitro but translational applications hampered by the high cost of production. We R6, a chemically-defined, albumin-free, low-transferrin culture medium, and MNC-RED, protocol differentiate peripheral mononuclear into enucleated erythroid that does not require any albumin or animal components. Erythropoiesis requires large amounts iron for hemoglobin synthesis. In all existing protocols, these needs are met increasing concentration holo-transferrin. This is necessary because transferrin recycling take place conditions. R6 provided differentiating erythroblasts small recombinant supplemented with FeIII-EDTA, an chelator allows cell culture. As result absence use low transferrin, production cultured using MNC-RED much less expensive than protocols. The should therefore help make many RBCs economically more feasible.
