Clean-Up Human Embryonic Stem Cell Lines Using Humanized Culture Condition.

摘要: Human embryonic stem cell (hESC) culture system has been changing conditions from conventional to xeno-free for therapeutic applications, and N-glycolylneuraminic acid (Neu5Gc) could be a useful indicator of xenogeneic contaminations in hESCs because human cells can no longer produce it genetically. We set up the humanized condition using commercially available materials two different adaptation methods: sequential or direct. SNUhES4 H1 hESC lines, previously established conditions, were maintained examined presence Neu5Gc. The showed same morphology character as those condition. Moreover, they negative Neu5Gc within passages without loss pluripotency. This study suggested that this method effectively cleanse bringing them one step closer being clinical-grade hESCs.
