摘要: Photodynamic treatment of the yeast Saccharomyces cerevisiae with singlet oxygen sensitizer toluidine blue and visible light leads to rapid oxidation ergosterol accumulation oxidized derivatives in plasma membrane. The predominant product accumulated was identified as 5α,6α-epoxy-(22E)-ergosta-8,22-dien-3β,7α-diol (8-DED). 9(11)-dehydroergosterol (DHE) a minor product. In heat inactivated cells is photooxidized epidioxide (EEP) DHE. Disrupted cell preparations S. cerevisiae convert EEP 8-DED, this activity abolished boiled control indicating presence membrane associated enzyme an isomerase activity. Yeast selectively mobilizes from intracellular sterol ester pool replenish level free during oxidation. following reaction pathway proposed: oxygen-mediated mainly formation EEP, which enzymatically rearranged 8-DED. Ergosterol 7-hydroperoxide, known ergosterol, formed at much lower rate decomposes give Changes physical properties are induced by depletion polar derivatives. Subsequent permeation photosensitizer through into events including impairment mitochondrial function inactivation.
