摘要: The ability of high density lipoproteins (HDL) to induce the clearance cholesteryl esters from cultured cells has been explored. Studies using J774 mouse macrophage cell line showed that these are not stimulated clear esterified cholesterol upon exposure HDL. This was observed over a wide range HDL concentrations (10 1000 micrograms/ml protein), and lack stimulation influenced by number factors relating preparation HDL, such as subfraction, varying extents lecithin:cholesterol acyltransferase modification, or heparin-Sepharose chromatography remove particles containing apo E. Neither method loading with nor physical state lipid droplets affected inability elicit clearance. In presence acyl CoA:cholesterol inhibitor, Sandoz 58-035, where level intracellular free generated, efflux only small fraction excess observed. were able efficiently in cholesterol-free apolipoprotein HDL/phospholipid particles, indicating have capacity cholesterol. Fu5AH hepatoma P388.D1 also failed response contrast, peritoneal macrophages cleared there fundamental differences between other types studied regard metabolism
