摘要: Biodegradable polyester polyhydroxyalkanoate (PHA) is a promising bioplastic material for industrial use as replacement petroleum-based plastics. PHA synthase PhaC forms an active dimer to polymerize acyl moieties from the substrate acyl-coenzyme A (CoA) into polymers. Here we present crystal structure of catalytic domain Chromobacterium sp. USM2, bound CoA. The reveals asymmetric dimer, in which one protomer adopts open conformation CoA, whereas other closed CoA-free form. stabilized by dimerization and enables accommodate CoA also create product egress path. molecule has its β-mercaptoethanolamine moiety extended site with terminal SH group close center Cys291, enabling formation reaction intermediate acylation Cys291.
