摘要: In the study of enzyme-catalysed mechanisms, it is often necessary to calculate upper limit enzyme reaction, which usually used as an important criterion identify whether assumed mechanism reasonable or not. Basically, in existing methods, there are two kinds models, Alberty-Hammes-Eigen model and Chou model, for calculating reaction. this paper, analysis comparison between these models made. It pointed out that magnitude van der Waals' binding energy substrate molecules will play a key role deciding significant difference not results calculated from models. Through such comparison, major protein outside active site molecule also becomes obvious: if very small, act like ‘wall’, blocking flow some site; while greater than 3 kT (where k Boltzmann constant T absolute temperature), behave ‘accelerator’, speeding up around molecule.
