摘要: The glucuronyl C5 epimerase (HSepi) is one of the modification enzymes involved in biosynthesis heparan sulfate (HS) and heparin, catalyzing epimerization D-glucuronic acid (GlcA) to L-iduronic (IdoA) at polymer level. IdoA critical for HS heparin interact with protein ligands, because its flexible conformation. Although enzyme recognizes both GlcA as substrates a reversible reaction hexuronic acids vitro, appears irreversible vivo. Targeted interruption gene, Glce, mice resulted neonatal lethality accompanied kidney agenesis, premature lung, skeletal malformations, demonstrating that single gene coded essential animal development. Elimination abnormal structure completely lacks residues. Loss 2-O-sulfation due lacking chains compensated by increased N- O-sulfation glucosamine Recombinant HSepi used generate HS/heparin related compounds having potential be therapeutic purposes.
