摘要: Peptidylarginine deiminase 4 (PAD4) is the only isoform of PADs located within cell nucleus, which has been known to be related several human diseases. In this work, we have proposed an electrochemical method for assay endogenous PAD4 activities as well studies inhibitors by making use supramolecular chemistry-assisted signal labeling. Specifically, peptide probes P1 and P2, separately contain cysteine residues tripeptides FGG (Phe-Gly-Gly), can self-assembled onto surface gold electrode silver nanoparticles, respectively. meantime, connected together through cucurbit[8]uril-mediated host–guest interaction. Nevertheless, after trypsin-catalyzed digestion, at N-terminal will removed from surface, thereby inhibiting connection P2.
