摘要: Limited data are available on the effects of stem cells in non-ischemic dilated cardiomyopathy (DCM). Since diffuse nature disease calls for a broad distribution cells, this study investigated scaffold-based delivery human induced pluripotent cell-derived cardiomyocytes (hiPS-CM) mouse model DCM. Nanofibrous scaffolds were produced using clinical grade atelocollagen which was electrospun and cross-linked under different conditions. As assessed by scanning electron microscopy shearwave elastography, optimum crosslinking conditions hiPS-CM colonization proved to be 10% concentration citric acid agent 150 min post-electrospinning baking. Acellular collagen first implanted both healthy mice those with DCM cardiac-specific invalidation serum response factor (SRF). Seven fourteen days after implantation, safety scaffold demonstrated echocardiography histological assessments. The subsequent step implantation seeded mice, cell-free as controls, revealed that heart function decreased controls while it remained stable treated mice. This pattern associated an increased number endothelial line greater vascularity scaffold. Moreover, lesser degree fibrosis consistent upregulation several genes involved extracellular matrix remodeling observed. These results support interest proposed stabilization outcome potential its use future.
