摘要: Abstract Unsaturated lipids exhibit different physiological significances due to the locations of carbon–carbon double bond (C=C). Identifying lipid isomers with mass-based methods remains challenging. Xia's group has been successfully employed Paterno–Buchi (PB) reaction, a photochemical reaction UV irradiation, coupled tandem mass spectrometry (MS/MS) identify and quantify unsaturated in complex mixtures. However, existing PB reagents possess certain demerits. In this regard, new reagent that is compatible various lipidomic analysis platforms must be screened. study, we comprehensively evaluated conditions screened benzophenone as reagent. Benzophenone possesses unique advantages, such relative high yield; products could readily distinguished from reacted based on added molecular weight (182 Da); does not affect appearance interval for reversed-phase column separation. Furthermore, optimized by using summarized formulas diagnostic ions according fragment rules. The proposed method implemented shotgun LC–MS lipidomics. To our best knowledge, work first report integrated lipidomics identification isomers.
