Photocoagulated human retinal pigment epithelial cells produce an inhibitor of vascular endothelial cell proliferation

摘要: Purpose. To determine if conditioned culture medium of photocoagulated human retinal pigment epithelial (RPE) cells contains inhibitors for the proliferation bovine aortic endothelial (BAEC) and (BREC) to characterize nature these inhibitory factors. Methods. Retinal grown confluence were (0.1 second, 50 μm, 350 mW) in serum-free medium. After 48 hours, (PC-CM) was removed, effects non-acid-treated transiently acid-treated samples determined on [ 3 H]-thymidine uptake by BAEC BREC. PC-CM also subjected size exclusion high-performance liquid chromatography (HPLC). Fractions analyzed growth before after transient acid treatment. Results. The addition (32% vol/vol) inhibited BREC 18.5% control value. With HPLC, activity recovered mainly a fraction whose apparent molecular 25 kd. treatment fractions, there appeared 100-kd inhibitor. Inhibitory neutralized pretreatment fractions with antiserum against transforming factor (TGF)-β2. Conclusions. Photocoagulated RPE secrete proliferation. Molecular immunologic properties correspond those TGF-β2. Invest Ophthalmol Vis Sci. 1995 ;36 :1686-1691.
