摘要: A comparative analysis of eight cytotoxicity assays [the 3T3 and normal human keratinocytes Neutral Red Uptake (NRU) assay, the primary rat hepatocytes, HepG2 MTT A.704, SH-SY5Y cells propidium iodide (PI) assay] included in several work packages EU Integrated Project ACuteTox, has been carried out. The aim was to evaluate whether originating from liver, kidney brain provided different vitro acute toxicity results, influence liver versus cell lines originated same tissue. Spearman rank correlation Hierarchical Cluster Analysis were performed based on IC50 (50% inhibitory concentrations for endpoint measured) values generated 57 chemicals. relatively large number neurotoxicants hepatotoxicants which allowed examine impact chemicals with specific tissue results. Our analyses confirmed similarity between NRU two hepatic systems related assays. type assay appears have greatest upon clustering result regardless origin used. information by differed that PI assay. This approach did not allow show but it does reveal effectiveness methodology choosing a testing program predicting e.g. oral humans.
