摘要: Clinical trials report improved overall survival following neoadjuvant chemoradiotherapy in patients undergoing surgery for esophageal adenocarcinoma, with a 10–15% improvement. MicroRNAs (miRNAs) are small noncoding RNAs that known to direct the behavior of cancers, including response treatment. We investigated ability miRNAs predict outcomes after chemoradiotherapy. Endoscopic biopsies from adenocarcinomas were obtained before and esophagectomy. miRNA levels measured using next generation sequencing compared pathological surgical resection, subsequent survival. ratios predicted identified by Lasso regression leave-one-out cross-validation. Association between ratio candidates relapse-free was assessed Kaplan–Meier analysis. Cox Harrell’s C analyses performed assess predictive performance miRNAs. Two (miR-4521/miR-340-5p miR-101-3p/miR-451a) found be associated Pretreatment expression these two ratios, pretreatment tumor differentiation, posttreatment AJCC histopathological grading, clearance/margins significant factors Multivariate analysis together pretherapy resulted risk prediction accuracy 85% (Harrell’s C), which comparable treatment grading (77%). miRNA-ratio biomarkers can used disease free esophagectomy adenocarcinoma.
