Prolonged hematopoiesis in a primate bone marrow culture system: characteristics of stem cell production and the hematopoietic microenvironment

摘要: Maintenance of myelopoiesis and pluripotential stem cell production for prolonged periods in vitro hitherto has been limited to mouse bone marrow culture. In an effort adapt the system use higher species, particularly human non-human primates, studies were undertaken using prosimian Tupaia glis (tree shrew). a number experiments duration sustained normal hematopoiesis observed cultures this following single inoculum 5 X 10(6)--10(7) cells, with or without addition fresh allogeneic exceeded 1 yr. Analysis suspension cells obtained by weekly demidepopulation such revealed CFU-C, differentiating neutrophils, basophils at high levels. Direct comparison murine indicated that both species complex series cellular interactions takes place within adherent environment marrow-derived endothelial macrophages, fat-containing cells. Certain functional ultrastructural features served distinguish from cultures, latter could be attributed regenerative capacity possessed its hematopoietic microenvironment. The availability primate culture should facilitate hematopoiesis, viral leukemogenesis, transplantation biology, which have more direct relevance man than provided existing system.
