摘要: Abstract Opioids have stimulatory as well the traditional inhibitory effects on neurotransmission, but underlying mechanisms are poorly understood. Here, Darren Smart and David Lambert review of opioids second messengers, including inositol (1,4,5)-trisphosphate (IP 3 ), protein kinase C (PKC), Ca 2+ , cAMP, propose that these coordinated changes at cellular level underlie facilitatory neurotransmission. The evidente for a possible role effects, particularly activation PKC by opioids, in development tolerance is also discussed.
