In situ detection of expression of the gus reporter gene in transgenic plants: ten years of blue genes

摘要: Among the methods now available to localize sites of gene expression in plant materials, reporter genes based on thegus (uidA) ofEscherichia coli, which encodes a β-glucuronidase (E.C. 3.2.1.31; GUS), have been most widely used during last ten years. The apparent simplicity histochemical GUS assay has major factor increase articles usinggus genes. However, over four years, there occasional reports expressing doubts concerning specificity observed localizations discrepancies between results obtained with histochemistry and immunocytochemistry and/orin situ hybridization. This brief review compares those various substrates for studies. Certain sources artefact are discussed, as limits that should be imposed interpretation at organ, tissue cell levels.
