A randomized clinical trial comparing radiation therapy v radiation therapy plus cis-dichlorodiammine platinum (II) in the treatment of locally advanced non-small cell lung cancer.

摘要: Cis-Dichlorodiammine platinum (II) (cis-DDP) was demonstrated to be a potentiator of radiation therapy (RT) in experimental tumor models and cultured cells. To assess the effectiveness combined modality treatment including RT weekly low-dose administration cis-DDP, from January 1986 June 1987, 95 patients with unresectable locally advanced non-small cell carcinoma lung (stage IIIa, b) were randomized for study. Fifty received alone at doses 50 Gy; 45 same plus cis-DDP 15 mg/m2 IV weekly. An overall response rate 50% 64% observed + group, respectively. No statistically significant differences detected regard median survival time (11 months v 16 cis-DDP) progression-free interval (7 arm 9 arm), but patterns first failure appeared affected by treatment. In fact, lower number intrathoracic relapses (12 23 arm). Toxicity mild feasibility this schedule must remarked. A better local control disease can obtained using as potentiator, true influence on length survival, optimal timing dosage are still evaluated further clinical trials.
