NS21: Re-defined and modified supplement B27 for neuronal cultures

摘要: In vitro culturing of primary neurons is a mainstay neurobiological research. Many these culture paradigms have taken advantage defined media rather than serum additives that contain undefined survival factors to facilitate experimental manipulations and interpretation the results. To in absence serum, supplements such as B27 are now widely used. However, commercially available exhibit large variability their capabilities support culture. We re-optimized modified earlier published formulations using 21 different ingredients (NS21). NS21 supports neuronal cultures high quality manifested by morphological characteristics, formation synapses, postsynaptic responses. Much B27/NS21 was due sources bovine albumin. Furthermore, we found holo-transferrin used preferable over apo-transferrin for cultures.
