Stimulation of arterial smooth muscle L-type calcium channels by hydrogen peroxide requires protein kinase C.

摘要: Changes in intracellular calcium regulate countless biological processes. In arterial smooth muscle, voltage-dependent L-type channels are major conduits for entry with the primary function being determination of diameter. Similarly, changes redox status, either discrete controlled or global pathological perturbations, also critical determinants cell function. We recently reported that muscle cells, local generation hydrogen peroxide leads to colocalized through channels. Here we extend our investigation into mechanisms linking influx by focusing on role protein kinase C (PKC). Our data indicate stimulation requires oxidant-dependent increases PKC catalytic activity. This effect is independent classical cofactor-dependent activation diacylglycerol. These provide additional experimental evidence supporting concept oxidative
