摘要: Publisher Summary This chapter focuses on contemporary approaches to the design of structurally defined peptides and proteins with special emphasis being given ways in which this approach has elucidated structural basis for function a variety natural molecules. When their proper biological milieu, assume fairly well-defined conformations, are responsible physical properties. The mechanisms by these molecules adopt three-dimensional structures depend size chemical compositions. There strategies protein peptide design. Those adopted problem will molecular characteristics desired target molecule. If one wishes create model small (e.g., pentapeptide hormone enkephalin), then it might be feasible an analog molecule that is locked into biologically active conformation. predetermined perhaps most ambitious goal, been attacked only recently. conformationally constrained powerful method elucidating conformation when bound its receptor. medium-sized beset many same problems encountered peptides. Medium-sized also very flexible tend lack conformations at room temperature aqueous solutions.
