摘要: The invention provides a method and materials for sorting polynucleotides with oligonucleotide tags. Oligonucleotide tags of the are capable hybridizing to complementary oligomeric compounds consisting subunits having enhanced binding strength specificity as compared natural oligonucleotides. Such referred herein "tag complements." Subunits tag complements may consist monomers non-natural nucleotide analogs, "antisense monomers" or they comprise oligomers lengths in range 3 6 nucleotides analogs thereof, including antisense monomers, being selected from minimally cross-hybridizing set. In such set, duplex made up an oligomer set complement any other contains at least two mismatches. Preferred include peptide nucleic acid nucleoside phosphoramidates 3'-NHP(O)(O--)O-5' linkage its adjacent nucleoside. An important aspect is use by specifically attached their on solid phase supports. This embodiment readily automated system manipulating polynucleotides, particularly useful large-scale parallel operations, DNA sequencing, mRNA fingerprinting, like, wherein many target segments single polynucleotide sequenced simultaneously.
