Curcumin: a potential therapeutic polyphenol, prevents noradrenaline-induced hypertrophy in rat cardiac myocytes

摘要: Objectives  This study was designed to evaluate the effect of curcumin on H9c2 cardiac cell line and primary rat myocytes, using purified noradrenaline as a hypertrophy-inducing agent. Methods  The concentration at which cells were treated determined by MTT (3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay. this safe dose in preventing noradrenaline-induced hypertrophy assessed biochemical analysis (estimating total protein content), molecular (using RT-PCR expression fetal genes like ANF), immunological (by determining nuclear localization GATA-4) electrophoretic mobility shift assay (EMSA; DNA binding activity GATA-4). Key findings  Curcumin 8 µm found suppress increase size, content enhanced marker gene (ANF) caused noradrenaline. Immunocytochemistry Western blot showed that suppressed transcription factor GATA-4 nucleus. It also reduced DNA-binding presence confirmed EMSA. Conclusions  These findings suggest reduces hypertrophic inhibiting GATA-4. Thus it has great anti-hypertrophic potential.
