摘要: Objectives. To evaluate long-term changes in acetylcholinesterase (AChE) activity CSF and blood following donepezil treatment relation to the concentration of cognition AD patients. Methods. or (or both) samples a total 104 patients with mild were used [MMSE score 23 ± 0.4; age 75 1 years (mean SEM); n = 53 for 51 plasma/red cell (RBC) samples]. The treated 5 10 mg/day clinically followed 2 years. RBC AChE activities measured by Ellman’s direct colorimetric assay. Protein levels two variants (“read-through” AChE-R synaptic AChE-S) determined an ELISA-like method. Results. plasma was dose-dependent (between 30 60 ng/mL 5-mg 10-mg group, respectively). 10 times lower than level showed dose- time-dependent kinetics. inhibition moderate (19–29%). AChE-S estimated 30–40% 45–55% group. Positive correlations observed between inhibition, increased protein variant MMSE examination. Patients high (≥45%) stabilized test result after up years, while significant decline (≤30%). Conclusions. An increase corresponded favored less cognitive deterioration.
