Histones, protamine, and polylysine but not poly(E:K) enhance transfection efficiency

摘要: Liposomal gene delivery has a great potential for the treatment of cancer and other human diseases. In this work we have investigated optimal conditions liposome-mediated transfer luciferase to erythroleukemia K562 cells. DDAB:DOPE liposomes were more efficient than lysyl-DOPE:DOPE (1:2) DDAB:cholesterol transfection. Total histones from bovine thymus, salmon sperm protamine, polylysine at an ratio 0.5 mg protein/mg DNA enhanced up 7-fold transfection efficiency plasmids; on contrary, synthetic polymer poly(E:K), containing glutamic acid lysine residues in random order 1:4, diminished expression. Transfection was nearly zero high histone:DNA by reversal charge particle negative positive leading:to its inability interact with cationic liposomes. The increase expression DNA-binding proteins might arise increased across cell membrane liposome-DNA-protein complex but also nuclear import DNA-protein because presence localization signals protein molecule used condensation.
